The Invitrogen™ Anza™ T4 PNK (polynucleotide kinase) Kit is used to perform 5-phosphorylation of DNA and oligonucleotides. T4 PNK exhibits 5 polynucleotide kinase and 3phosphatase activity, catalyzing the transfer of the terminal phosphate of ATP to a 5 hydroxyl group of a nucleic acid.

When preparing DNA for the ligation step in cloning, either the insert DNA or the vector DNA should contain a 5phosphate. Often PCR-amplified DNA is treated with T4 PNK to phosphorylate the 5 end for subsequent cloning ligation.
