Invitrogen™ Anza™ T4 PNK Kit

The Invitrogen™ Anza™ T4 PNK (polynucleotide kinase) Kit is used to perform 5′-phosphorylation of DNA and oligonucleotides.
T4 PNK exhibits both 5′ polynucleotide kinase and 3′ phosphatase activity, catalyzing the transfer of the terminal phosphate of ATP to a 5′ hydroxyl group of a nucleic acid.

Application

When preparing DNA for the ligation step in cloning, either the insert DNA or the vector DNA should contain a 5′ phosphate.
Often, PCR-amplified DNA is treated with T4 PNK to phosphorylate the 5′ end for subsequent cloning ligation.
