Description:
The CD45-2B11 monoclonal antibody reacts with human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except erythrocytes and platelets. Its cytoplasmic domain possesses tyrosine phosphatase activity, crucial for lymphocyte proliferation and differentiation. The 2B11 antibody is useful for identifying normal and neoplastic lymphoid cells.
Applications Reported:
Suitable for flow cytometric analysis, immunoblotting, immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections, and immunocytochemistry.
Applications Tested:
- IHC (Paraffin): ≤10 µg/mL (high pH antigen retrieval)
- WB: 5 µg/mL
- Flow Cytometry: 0.25 µg/test on human peripheral blood cells
A test is defined as the amount (µg) of antibody used to stain a cell sample in 100 µL final volume. Cell number may vary from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information
CD45 (LCA, Leukocyte Common Antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, constituting ~10% of lymphocyte surface proteins. It is absent in non-hematopoietic tissues and cell lines. CD45 is essential for lymphocyte development and antigen signaling, regulating Src-family kinases and JAK kinase pathways. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing transmembrane and cytoplasmic regions. CD45 plays roles in T/B-cell receptor signaling, cytokine signaling regulation, integrin-mediated survival, and apoptosis modulation.
