Target Information
TLR4는 Toll-like receptor (TLR) 계열의 구성원으로, 병원체 인식과 선천면역 활성화에 핵심적인 역할을 합니다. TLR은 초파리부터 인간까지 고도로 보존되어 있으며 구조적, 기능적 유사성을 공유합니다.
이 수용체는 주로 태반과 백혈구의 myelomonocytic 아형에서 많이 발현됩니다. 포유류 세포는 LPS에 반응하여 TLR4를 활성화하며, CD14, LY96과 함께 LPS 수용체 복합체를 형성해 신호전달 과정을 매개합니다.
TLR4는 병원체 관련 분자 패턴(PAMPs)을 인식하여 면역 반응에 필요한 사이토카인 생성을 유도합니다. TLR4 유전자의 변이는 LPS 반응성의 차이와 관련이 있으며, 여러 전사 변이가 보고되어 있습니다.
TLR4는 말초혈 단핵구와 일부 B세포, 인간 태반에서 발현되며, TLR4 결핍 생쥐는 LPS에 대한 반응이 감소하고 그람음성균 패혈증에 취약함이 보고되었습니다.
