Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineages and activated mature lymphocytes. It associates with CD122 and CD132 to form the high-affinity IL-2 receptor complex.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤ 0.05 µg/test (100 µL volume). Optimize by titration for best results.

• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology for engineered fluorescent signatures minimizing spectral spillover.

Target Information

CD25 (IL2RA) encodes the interleukin-2 receptor alpha chain, part of a heterotrimeric receptor complex (IL2RA, IL2RB, IL2RG). This receptor mediates IL-2-dependent proliferation of T and B lymphocytes. IL2RA is critical for immune regulation and lymphocyte activation. Mutations can lead to immune dysregulation, such as ulcerative colitis-like disease in mice.
