Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon stress, p53 activates transcription of target genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 contains:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a central role in preventing proliferation of damaged cells, acting as a key anticancer factor. It is also implicated in aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP).

In normal cells, p53 levels are low, but in transformed cell lines, its expression is elevated and contributes to malignancy. Mutations in TP53 disrupt DNA binding and abolish tumor suppressor activity. Both somatic and germline mutations (e.g., Li-Fraumeni syndrome) are associated with human cancers.
