Target Information
TRAIL (TNF-related apoptosis-inducing ligand) is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells while sparing normal cells, despite widespread expression in normal tissues. TRAIL interacts with several TNF receptor superfamily members, including:
- TNFRSF10A/TRAILR1 (DR4)
- TNFRSF10B/TRAILR2 (DR5)
- TNFRSF10C/TRAILR3 (DcR1)
- TNFRSF10D/TRAILR4 (DcR2)
- TNFRSF11B/OPG
Binding to decoy receptors (TRAILR3, TRAILR4, OPG) modulates TRAIL activity by preventing apoptosis induction. TRAIL signaling triggers activation of MAPK8/JNK, caspase 8, and caspase 3, leading to programmed cell death and NF-κB activation in various tissues.
