Target Information

This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant).
These chemokines belong to the CXC subfamily, where a single amino acid separates the first two of four conserved cysteine residues.

Binding of these chemokines induces cellular responses involved in leukocyte trafficking, integrin activation, cytoskeletal rearrangement, and chemotactic migration.
Inhibition by Bordetella pertussis toxin suggests coupling with Gi-type G proteins.

Signal transduction mechanisms are thought to be similar to those of other chemokine receptors, involving phosphorylation-dependent receptor internalization and recycling.
CXCR3 is highly expressed in effector/memory T cells and in inflamed tissues, contributing to inflammatory cell recruitment.

This receptor is a potential target for small-molecule antagonists aimed at treating inflammatory diseases.
Multiple transcript variants encoding different isoforms have been identified.
