Target Information
XBP1 encodes a transcription factor that regulates MHC class II genes by binding to a promoter element referred to as an X box. This gene product is a bZIP protein, also identified as a cellular transcription factor binding to an enhancer in the T cell leukemia virus type 1 promoter. It may increase expression of viral proteins by acting as the DNA-binding partner of a viral transactivator.
When unfolded proteins accumulate in the endoplasmic reticulum (ER), XBP1 mRNA is processed to an active form by an unconventional splicing mechanism mediated by IRE1. The spliced mRNA loses 26 nucleotides, producing the functionally active XBP1(S) isoform. The unspliced form, XBP1(U), acts as a negative feedback regulator of XBP1(S), shutting off transcription of target genes during ER stress recovery. A pseudogene of XBP1 is localized to chromosome 5.
