Target Information
Apoptosis is related to many diseases and developmental processes. Caspase-9 plays a central role in cell death induced by various apoptosis activators. Upon release from mitochondria, cytochrome c binds to Apaf-1 to form an apoptosome that activates procaspase-9. Activated caspase-9 then cleaves and activates effector caspases (caspase-3, -6, and -7), which mediate the proteolytic cleavage of key proteins during apoptosis. PHAP proteins have been identified as important regulators of mitochondrial apoptosis, facilitating apoptosome-mediated caspase-9 activation and stimulating the mitochondrial apoptotic pathway. PHAP also opposes Ras- and Myc-mediated cell transformation.
