Target Information
SOAT1 (sterol O-acyltransferase-1), also known as ACAT1, is a homotetrameric enzyme that catalyzes the formation of cholesterol esters from cholesterol and long-chain fatty acyl-coenzyme A (acyl-CoA). The human SOAT1 gene maps to chromosome 1 and encodes a protein localized to the endoplasmic reticulum (ER) in tissues such as liver, kidney, adrenal glands, and macrophages.
SOAT1 plays a key role in cellular cholesterol homeostasis, foam cell formation, and atherosclerosis progression. Several SOAT inhibitors have been developed for potential therapeutic use in atherosclerosis.
SOAT2 (sterol O-acyltransferase-2), also known as ACAT2, participates in lipoprotein assembly and cholesterol esterification in mammalian cells. It is an integral ER membrane protein of intestinal cells, and SOAT2 deficiency is associated with severe mental retardation and hypotonus.
