Target Information

The p38 MAPK cascade regulates various cellular responses to stress, inflammation, and other signals. p38 MAPK is relatively inactive in its nonphosphorylated form and becomes rapidly activated by dual phosphorylation at the Thr-Gly-Tyr motif.
There are four isoforms of p38 MAPK, differing in tissue expression and affinity for upstream activators and downstream effectors.
Upon exposure to stimuli such as TNF, IL-1, or heat shock, MAPK kinase-3/6 phosphorylates Thr180 and Tyr182 residues in p38 MAPK, leading to activation.
Activated phospho-p38 MAPK subsequently phosphorylates transcription factors such as ATF-2, CHOP-1, and MEF-2.
