Target Information
ERN1 (endoplasmic reticulum to nucleus signaling 1) is a serine/threonine kinase and endoribonuclease that serves as the sensor of unfolded proteins in the endoplasmic reticulum (ER). It initiates signal transduction from the ER to the nucleus by splicing XBP1 mRNA, converting it into a potent transcriptional activator for the unfolded protein response. ERN1 plays a critical role in extraembryonic cells essential for fetal viability. IRE1 is vital under stress conditions that lead to unfolded protein accumulation in the ER. Depletion of IRE1p through dominant negative expression induces apoptosis under such stress conditions.
