Applied Biosystems™ PureQuant B Cell Assay

The Applied Biosystems PureQuant assays are a family of TaqMan qPCR-based assays designed to accurately identify and quantify specific immune cell types within a mixed cell population. These assays measure genomic DNA methylation at specific sites that serve as unique identifiers for each cell type.

Each 96-well plate can accommodate up to seven test samples, and results are provided as a percentage of the target cell type out of total input cells using a free analysis tool.

Assay Principle

The PureQuant B Cell Assay utilizes the LRP5 gene as an epigenetic marker for B cells.

Kit Components

• Calibrator: Corrects for variation in bisulfite conversion efficiency. Plasmid-based calibrator includes genomic target regions and GAPDH, used to estimate assay-specific bisulfite efficiency factor.
• Reference: Genomic DNA from pooled healthy donors used to verify assay performance. Reference values reflect expected target cell type ranges.
• Standards: Six dilutions spanning a 4-log dynamic range (30–31,250 copies) for determining unknown copy numbers using standard curves.
