This product is diluted and provided in a ready-to-use formulation.
A recommended positive control tissue is pituitary, though other tissues may also be suitable.
The primary antibody is intended for laboratory professional use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue stained in manual qualitative immunohistochemistry (IHC) testing. It is used after the primary diagnosis of a tumor has been made by conventional histopathology using non-immunological histochemical stains.
ACTH (Corticotropin) is a 39-amino-acid active peptide produced by the anterior pituitary. This monoclonal antibody is specific to CLIP (aa 25–39 of ACTH) and does not react with Synacthen (aa 1–24 of ACTH).
POMC (pro-opiomelanocortin) is a 267-amino-acid precursor hormone that undergoes tissue-specific posttranslational processing by convertases, producing multiple hormone chains including ACTH, MSH variants, CLIP, lipotropins, and endorphins.
ACTH is also produced by immune cells (T-cells, B-cells, macrophages) in response to stress stimuli. Anti-ACTH is a useful marker for classifying pituitary tumors and studying pituitary disease. It reacts with ACTH-producing cells (corticotrophs) and may also react with other tumors (e.g., small cell carcinomas of the lung) associated with paraneoplastic syndromes secreting ACTH.
Usage Notes
- Used with formalin-fixed, paraffin-embedded sections.
- Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
- IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, with washing steps between each stage.
- Results are visualized via light microscopy for differential diagnosis.
Controls
- A positive tissue control must be run with every staining procedure.
- Should include both positive and negative staining regions.
- Fresh autopsy/biopsy/surgical specimens fixed and embedded similarly to patient samples are recommended.
- Weakly positive tissues are ideal for detecting subtle changes in antibody sensitivity or reagent stability.
- Negative control tissues (internal or external) should be used according to institutional guidelines to confirm absence of non-specific staining.
Target Information
ATCH (Adrenocorticotropic Hormone) plays a major role in stimulating the adrenal cortex.
It is formed by cleavage of proopiomelanocortin (POMC), which also yields MSH and beta-endorphin.
ATCH secretion is triggered by corticotropin-releasing hormone from the hypothalamus and stimulates glucocorticoid (e.g., cortisol) secretion from the adrenal cortex, with minimal effect on mineralocorticoid (aldosterone) production.
