Target Information
Nerve growth factor (NGF) influences the survival and differentiation of various neuronal and non-neuronal cells. Its biological activity is mediated by binding to specific cell surface receptors, existing as high- and low-affinity forms. Only the high-affinity receptor mediates signal transduction. Cross-linking studies with 125I-NGF have identified receptor complexes of Mr 158,000 (high affinity) and Mr 100,000 (low affinity) in rat PC12 cells.
Full-length cDNA clones for human and rat NGF receptor genes show over 90% homology. NGF is crucial for nerve growth stimulation, regulating growth and differentiation of sympathetic and sensory neurons. Mutations in the NGF gene are linked to hereditary sensory and autonomic neuropathy type 5 (HSAN5), and dysregulation of NGF expression is associated with allergic rhinitis.
