Target Information

Epithelial sodium channels (ENaC) are amiloride-sensitive members of the degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels. These channels share structural similarity with two short intracellular termini, two membrane-spanning segments, and a large extracellular loop containing a conserved cysteine-rich region.
ENaC isoforms (alpha, beta, gamma) are expressed in kidney, lung, and colon, playing essential roles in trans-epithelial sodium and fluid balance. In the kidney, ENaC mediates aldosterone-dependent sodium reabsorption in the distal nephron, contributing to blood pressure regulation.
ENaC function is regulated partly through interaction with the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel.
Gain-of-function mutations in β- or γ-ENaC can lead to severe arterial hypertension (Liddle’s syndrome), while loss-of-function mutations in α- or β-ENaC cause pseudohypoaldosteronism type 1 (PHA-1).
