Juvenile nephronophthisis type 1은 NPHP1 유전자의 변이에 의해 발생하며, NPHP1은 nephrocystin 단백질을 암호화합니다. Nephrocystin은 p130Cas (BCAR1), PTK2B (Pyk2), tensin 등과 상호작용하여 신호 전달 경로에 관여합니다. 이 단백질은 세포 간 접촉 부위 근처에 위치하며, nephrocystin 발현은 Pyk2의 Tyr402 인산화 및 ERK1/2 활성화를 유도합니다. 또한, Nephrocystin은 진화적으로 보존된 SH3 도메인을 포함합니다.
