Target Information

Varp (Vps9 Domain and Ankyrin-Repeat-Containing Protein) may function as a guanine exchange factor (GEF) for Rab21, Rab32, and Rab38, regulating endosome dynamics. It may control VAMP7 participation in membrane fusion events by inhibiting SNARE complex formation.
Varp is involved in melanosomal distribution of TYRP1 in melanocytes and neurite growth regulation. It may also participate in Golgi sorting and transport of VAMP7 vesicles to the cell surface, interacting with kinesin heavy chain isoform 5, GOLGA4, RAB21, and MACF1.
Additionally, Varp is required for colocalization of VAMP7 and Rab21 and regulates GLUT1 endosome-to-plasma membrane trafficking in association with VPS29. It ensures proper trafficking of melanogenic enzymes TYR, TYRP1, and DCT/TYRP2 to melanosomes.
