Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except mature erythrocytes and platelets. Its cytoplasmic domain has tyrosine phosphatase activity crucial for lymphocyte activation.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse bone marrow cells (≤ 0.4 µg/test).
A test is defined as the amount of antibody that stains a cell sample in 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test. Titration is recommended for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample or per 100 µL of cell sample for single-color controls.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, accounting for ~10% of lymphocyte surface proteins. It is absent on non-hematopoietic cells and tissues. CD45 regulates Src-family kinases, cytokine receptor signaling, and lymphocyte activation. Multiple isoforms arise through alternative splicing, influencing expression and membrane localization. CD45 also modulates integrin-mediated signaling, apoptosis, and immune regulation.
