Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and mediate receptor internalization via ubiquitin pathways.
CD33 is expressed on myelomonocytic lineage cells and binds sialic acid residues on cell surface glycans. It serves as a therapeutic target for acute myeloid leukemia and is found on granulocyte and macrophage precursors but not on pluripotent stem cells.
CD33 is also expressed on peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
