Target Information
This gene generates several transcript variants that differ in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode isoforms functioning as inhibitors of CDK4 kinase. Another transcript contains an alternate open reading frame (ARF) that encodes a structurally unrelated protein, functioning as a stabilizer of the tumor suppressor protein p53 by interacting with and sequestering the E3 ubiquitin-protein ligase MDM2. Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product contribute to G1 cell cycle control through CDK4 and p53 regulation. This gene is frequently mutated or deleted in various tumors and is recognized as an important tumor suppressor gene.
