Target Information
Component of the adaptor protein complex 2 (AP-2). Adaptor protein complexes function in protein transport via vesicles in various membrane traffic pathways. These complexes act as vesicle coat components involved in cargo selection and vesicle formation. AP-2 is essential for clathrin-dependent endocytosis, where cargo proteins are incorporated into clathrin-coated vesicles (CCVs) destined for fusion with early endosomes.
The clathrin lattice serves as a mechanical scaffold but cannot directly bind to membrane components. Clathrin-associated adaptor protein (AP) complexes can bind to both clathrin and membrane proteins and lipids, acting as major clathrin adaptors contributing to CCV formation. AP-2 also serves as a cargo receptor to selectively sort membrane proteins involved in receptor-mediated endocytosis and may play a role in synaptic vesicle recycling.
AP-2 recognizes Y-X-X-[FILMV] (Y-X-X-Phi) and [ED]-X-X-X-L-[LI] endocytosis signal motifs within cytosolic tails of transmembrane cargo molecules. It may also participate in maintaining normal post-endocytic trafficking through the ARF6-regulated, non-clathrin pathway. The AP-2 alpha and sigma subunits contribute to recognition of the [ED]-X-X-X-L-[LI] motif and may also be involved in extracellular calcium homeostasis.
