Product Specific Information

The DX5 monoclonal antibody reacts with CD49b, an antigen expressed on a majority of mouse natural killer (NK) cells and a subset of T cells. DX5 recognizes all tested mouse strains including BALB/c, C57BL/6, C3H, CBA, DBA, AKR, SJL, and 129.
Simultaneous staining of C57BL/6 spleen cells with anti-NK1.1 mAb (PK136) and DX5 shows coexpression of both markers by most cells, with small populations of DX5⁺PK136⁻ and DX5⁻PK136⁺ cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes; can be used at ≤0.25 µg per test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

Target Information

CD49b (Integrin alpha 2) is a member of the integrin family, forming a heterodimeric complex with CD29 (Integrin beta 1) to create VLA-2 (integrin α2β1). This complex serves as an extracellular receptor for laminin, collagen, and fibronectin, activating intracellular signaling pathways.
It plays roles in VEGF-induced angiogenesis, adhesion, and lymphocyte activation. CD49b is expressed by NK cells, NK-T cells, monocytes, platelets, epithelial cells, and subsets of T and B cells, including CD4⁺ Tr1 regulatory cells co-expressing LAG-3.
