Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. CD20 expression coincides with surface IgM and regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 associates with lipid rafts, and its interaction depends on extracellular triggering and BCR aggregation. Upon receptor ligation, CD20 and BCR colocalize briefly before BCR endocytosis, while CD20 remains on the cell surface. CD20 is a key therapeutic target for antibody-mediated B-cell depletion, as it is highly expressed on B-cell malignancies and remains stable on the plasma membrane after antibody treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
