Background
Insulin-like growth factor-binding protein 7 (IGFBP-7)는 당화된 분비 단백질로, 세 가지 주요 도메인(N-terminal IGFBP domain, Kazal-type serine proteinase inhibitor domain, C-terminal immunoglobulin-like C2-type domain)을 포함합니다. 인간과 마우스 IGFBP-7은 94%의 아미노산 서열 일치를 가지며, 다양한 정상 조직과 암세포에서 발현됩니다.
IGFBP-7은 이차 림프 조직의 혈관 내 고내피 venule(HEV)에 풍부하게 존재하며, IGF 및 인슐린과 낮은 친화도로 결합합니다. IGF 및 인슐린의 세포 증식 작용을 강화하며, IGF/인슐린 비의존적 기능도 수행합니다. 또한 heparan sulfate proteoglycan, type IV collagen, 특정 chemokine과 상호작용하여 약한 세포 부착을 지원하고, collagen 위에서 세포 확산을 촉진하며, 강력한 혈관 확장 인자인 PGI2 생성을 유도합니다. 종양 세포 성장 조절 및 혈관 신생(angiogenesis)에 관여하는 것으로 알려져 있습니다.
