배경 정보
α-Galactosidase A는 glycosyl hydrolase 27 family에 속하는 호모다이머성 당단백질로, 리소좀 효소로 작용합니다. Fabry 질환 진단을 받은 환자에게 장기적인 효소 대체 요법으로 사용됩니다. 이 효소는 glycolipids 및 glycoproteins의 말단 α-galactosyl기를 가수분해하며, melibiose를 galactose와 glucose로 분해합니다.
GLA 유전자의 결함은 Fabry disease(FD)의 원인이며, 이는 희귀 X-linked sphingolipidosis로 여러 조직에서 glycolipid가 축적되는 질환입니다. FD 환자는 globotriaosylceramide(Gb3) 및 관련 glycosphingolipids가 혈장과 세포 리소좀에 축적되며, 안구 침착, 열성 발작, 사지의 작열통을 나타낼 수 있습니다. 사망은 신부전, 심혈관 또는 뇌혈관 합병증으로 이어질 수 있습니다.
