Background
Trophoblast glycoprotein (TPBG, also known as 5T4) belongs to the LRR protein family. The LRR N-terminal cap (LRRNT) and LRR1 domains are essential for inhibiting the Wnt signaling pathway. TPBG may act as a Wnt/β-catenin signaling inhibitor by indirectly interacting with LRP6 and blocking Wnt3a-dependent LRP6 internalization. It is expressed by all trophoblast types from as early as 9 weeks of development and, in adults, is limited to certain epithelial cells but found on various carcinomas.
