Background
Protein tyrosine phosphatase, receptor type C (CD45, PTPRC)는 protein tyrosine phosphatase (PTP) 계열에 속하며, 세포 증식, 분화, 유사분열 및 암화 과정 등 다양한 세포 기능을 조절하는 신호전달 분자입니다.
CD45는 모든 핵을 가진 조혈계 세포에서 풍부하게 발현되는 180–220 kDa 크기의 당단백질로, 세포 유형, 발달 단계 및 항원 노출에 따라 다양한 isoform을 가집니다.
특히 T 세포에서 TCR 신호 전달 임계값을 조절하며, 면역 시냅스(IS) 내외로 이동하며 galectin lattice 및 actin cytoskeleton과의 상호작용에 의해 위치가 조절됩니다.
CD45는 IS 내에서 src family kinase인 LCK를 탈인산화하여 음성 조절하며, 다른 백혈구에서는 분화, 사이토카인 분비 및 생존에 관여합니다.
CD45 결손은 심각한 면역결핍을 초래하며, 점 돌연변이는 자가면역 질환과 관련될 수 있습니다.
