Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 up-regulation leads to the activation of genes such as p21waf1, resulting in G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization of p53. Phosphorylation at Ser392 is linked to tumor formation. p53 plays roles in tumor suppression, aging, oxidative stress response, and neural function related to learning and memory.

Mutations in the TP53 gene are common in various human cancers, leading to loss of tumor suppressor activity. Germline mutations are also associated with Li-Fraumeni syndrome.
