Product Specific Information

The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, which associates with the B cell receptor (BCR). Signaling through CD19 induces tyrosine phosphorylation, calcium flux, and B cell proliferation.
Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Use in flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails prior to combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and certain myeloid leukemia cells. CD19 acts as a signal-amplifying coreceptor for BCR, regulating B cell proliferation and differentiation. Mutations cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
