Target Information
The BCKDHA gene encodes the E1-alpha subunit of the branched-chain alpha-keto acid dehydrogenase (BCKD) complex.
This complex consists of three catalytic components:
- A heterotetrameric (alpha2-beta2) branched-chain alpha-keto acid decarboxylase (E1)
- A homo-24-meric dihydrolipoyl transacylase
- A homodimeric dihydrolipoamide dehydrogenase
E1 is a thiamine pyrophosphate (TPP)-dependent enzyme, catalyzing an irreversible reaction that represents the first committed step in branched-chain amino acid (BCAA) oxidation. The BCKDHB gene encodes the beta subunit of E1. The complex also includes two regulatory enzymes: a kinase and a phosphorylase.
