Target Information
MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating p53 stability. It is located on chromosome 12q and undergoes phosphorylation by Akt at Ser166 and Ser186, enhancing interaction with p300 and promoting p53 ubiquitination and degradation. MDM2 also inhibits p53 transactivation and can inactivate p53 through E3 ubiquitin ligase activity. Overexpression of MDM2 is associated with various human cancers, including soft tissue carcinomas and breast cancer. It also interacts with proteins such as retinoblastoma 1 and ribosomal protein L5, contributing to apoptosis and cell cycle regulation. More than 40 alternatively spliced transcript variants have been identified in tumor and normal tissues.
