IFI16 (gamma-interferon-inducible protein 16)은 이중가닥 DNA에 결합하며, supercoiled 및 cruciform DNA 구조에 선호적으로 결합합니다. 전사 조절 및 억제 인자로 작용하며, 혈액세포 분화 조절, 세포주기 조절 인자(p53/TP53, RB 단백질 등) 기능 조절, 선천적 면역반응에서의 역할을 수행합니다. 또한, 바이러스 dsDNA 인식 후 TMEM173/STING을 모집하여 IFN-β 유도에 관여하며, HSV-1 감염 시 IFR3 신호 경로 활성화 및 바이러스 유전자 발현 억제에 기여합니다. IFI16은 유방암에서 ESR1 발현의 MTA1 매개 후성유전적 조절에도 관련됩니다.
