Product Specific Information

The 16-10A1 monoclonal antibody reacts with mouse CD80 (B7-1), a 55 kDa member of the Ig superfamily. CD80 is expressed by macrophages, dendritic cells, and activated B cells, as well as activated T cells. It binds to T cell surface antigens CD28 and CD152 (CTLA-4), playing a crucial role in T-B cell communication and activation.

• Applications Tested: Tested by flow cytometric analysis of stimulated mouse splenocytes (≤ 0.06 µg/test)
• Recommended Conditions: Final volume 100 µL, cell number 10⁵–10⁸ cells/test; titrate for optimal performance

Fluorochrome Information:
Super Bright 436 is excited with the violet laser (405 nm) and emits at 436 nm. Recommended detection with a 450/50 bandpass filter or equivalent.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD80 (B7-1) and CD86 (B7-2) are ligands for T cell costimulatory molecule CD28 and inhibitory receptor CTLA-4 (CD152). Both are expressed on antigen-presenting cells and are essential for T cell activation. CD80 is rapidly induced on activated B cells, EBV-transformed and Burkitt lymphoma cell lines, freshly isolated follicular B lymphoma cells, T cells, and monocytes. It regulates T lymphocytes via binding to CD28 and CTLA4.

Diseases associated with CD80 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
