Target Information

Poly(ADP-ribosyl)ation is an immediate DNA-damage-dependent post-translational modification of histones and other nuclear proteins that contributes to the survival of injured proliferating cells. Poly(ADP-ribose) polymerases (PARPs) constitute a family of 18 proteins encoded by different genes and sharing a conserved catalytic domain. PARP-1 (113 kDa) and PARP-2 (62 kDa) are the main enzymes whose catalytic activity is stimulated by DNA strand breaks. The expanding family of PARPs extends the role of poly(ADP-ribosyl)ation to various aspects of cell biology, including proliferation and cell death. Some members interact with each other, share partners and subcellular localizations, suggesting fine regulation of this post-translational modification.
