Target Information

ROCK1 is a serine/threonine kinase that regulates cell differentiation and migration. The Rho/ROCK pathway is involved in the progression of various human cancers. ROCK1 has been identified as a Caspase-3 substrate. It contributes to myosin light chain (MLC) activation through direct phosphorylation of MLC and inhibition of MLC phosphatase. This process increases cellular contractility, which is essential for apoptotic membrane blebbing.

ROCK1 consists of an amino-terminal kinase domain and a cysteine/histidine-rich C-terminal inhibitory domain within a pleckstrin-homology region, connected by a variable region containing the Rho-binding domain. During apoptosis, caspase-3 cleaves ROCK1 at a conserved DETD1113/G sequence, removing the inhibitory domain and resulting in constitutive kinase activity. This cleavage induces MLC phosphorylation and membrane blebbing, key events in the execution phase of apoptosis.
