Target Information
Lipolytic enzymes are essential for mobilizing fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis can affect energy homeostasis and contribute to obesity and insulin resistance.
Adipose triglyceride lipase (ATGL) catalyzes the initial step in triglyceride hydrolysis and is highly expressed in adipose tissue of mice and humans. It shows high substrate specificity for triacylglycerol and is associated with lipid droplets. Inhibition of ATGL significantly decreases total adipose acyl-hydrolase activity.
ATGL and hormone-sensitive lipase (HSL) work together to catabolize stored triglycerides in mammalian adipose tissue.
