Target Information
The expression pattern of the Prox1 homeobox gene suggests a role in various embryonic tissues, including the lens. Analysis of mRNA reveals that Prox1 mRNA is present in many human tissues, with the highest level in the lens. Homozygous Prox1- mice die at midgestation due to multiple developmental defects, including lens malformation. Prox1 inactivation results in abnormal cell proliferation, reduced expression of cell cycle inhibitors (Cdkn1b, Cdkn1c), misexpression of E-cadherin, and excessive apoptosis, leading to a hollow lens phenotype.
Prox1 is expressed in a subpopulation of endothelial cells that form the lymphatic system through budding and sprouting. It is a specific and essential regulator of lymphatic system development. In mice, Prox1 is also required for hepatocyte migration; its loss leads to a smaller liver with fewer clustered hepatocytes. In the embryonic mouse retina, Prox1 regulates progenitor cell cycle exit and differentiation. Cells lacking Prox1 continue dividing, whereas ectopic expression promotes cell cycle exit. Thus, Prox1 is a key regulator of progenitor-cell proliferation and cell-fate determination in the vertebrate retina.
