Target Information
Ago proteins are broadly expressed in somatic cells, associate with miRNAs, and are key actors in RNA silencing pathways. Among them, Ago2 uniquely exhibits endonucleolytic “Slicer” activity, enabling miRNA-directed cleavage of target mRNA when base-pairing is perfect. With partial complementarity, Ago2 represses translation instead of cleavage. It plays a non-redundant role in RNA interference, translation repression, and heterochromatinization. As a core component of the RISC complex, AGO2 catalyzes degradation of target mRNAs guided by siRNAs or miRNAs and also regulates miRNA maturation. Overexpression of Ago2 is linked to tumor growth and cancer prognosis. In mice, Ago2 is essential for embryonic development and regulation of B-lymphoid and erythroid lineages.
