Product Specific Information

The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein belonging to the Siglec family. It is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human lysed whole blood.
Use at 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer:

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample (regardless of number of NovaFluors)
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information

CD33 is a transmembrane protein of the Siglec family containing ITIM motifs that recruit SHP-1 and SHP-2 phosphatases. It is expressed on myelomonocytic lineage cells and binds sialic acid residues on cell surfaces.
CD33 is a therapeutic target for acute myeloid leukemia and serves as a marker for distinguishing myelogenous leukemia from lymphoid or erythroid leukemias.
It is present on granulocyte and macrophage precursors but absent on pluripotent stem cells.
Associated diseases include gallbladder lymphoma and extracutaneous mastocytoma.
