Target Information
The Ras superfamily of GTPases—including Ras, Rho/Rac, Sar, Rab, ARF, and Ran subfamilies—regulates diverse cellular processes such as cytoskeletal rearrangement, nuclear signaling, and cell growth. These GTPases act as molecular switches toggling between active (GTP-bound) and inactive (GDP-bound) states. Activation is mediated by guanine nucleotide exchange factors (GEFs).
Dbl-related proteins represent a large family of structurally related GEFs that act on specific Ras family members, including FGD1, Lsc, RhoGEF p115, Lfc, Lbc, and Brx. Among these, Lsc, Lbc, and Lfc share sequence homology and exhibit exchange activity toward Rho family GTPases. RhoGEF p115 specifically catalyzes GEF activity for Rho but not Rac, Cdc42, or Ras.
