Product Specific Information

The RA3-6B2 monoclonal antibody reacts with the exon A-restricted isoform of mouse CD45 (220 kDa). CD45R/B220 is primarily expressed on B cell lineage from early Pro-B to mature B cells. Some activated T cells, LAK cells, NK progenitors, and mutant T cells (lpr/lpr) also express this antigen.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse splenocytes at ≤0.1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before mixing with cells.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing engineered nucleic acid dye structures to minimize spillover and spread.

Target Information

CD45R (PTPRC) is a member of the protein tyrosine phosphatase (PTP) family involved in regulating cell growth, differentiation, mitotic cycle, and oncogenic transformation.
It is specifically expressed in hematopoietic cells and is essential for T- and B-cell receptor signaling.
CD45 glycoprotein exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
It plays key roles in:

• Lymphocyte development and antigen signaling
• Regulation of Src-family kinases
• Integrin-mediated signal transduction
• DNA fragmentation during apoptosis
