Product Specific Information

The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through its binding to hyaluronate, an extracellular matrix component.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse bone marrow cells and splenocytes.
Recommended ≤0.6 µg per 100 µL test (10⁵–10⁸ cells/test). Optimize by titration for best performance.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD44 is a 100 kDa type I transmembrane glycoprotein broadly expressed on human leukocytes, brain white matter, and epithelial cells. It serves as a receptor for hyaluronic acid (HA) and mediates cell-cell interactions, adhesion, and migration. CD44 participates in lymphocyte activation, recirculation, and homing, and its expression can increase in some carcinomas, potentially correlating with metastatic potential.

Alternative splicing of the CD44 gene produces multiple isoforms with distinct functions. CD44 dysfunction is associated with diseases such as superficial keratitis and lichen sclerosus, and may contribute to tumor metastasis formation.
