Target Information
STK4 (MST1) is a serine/threonine kinase involved in apoptosis. It induces chromatin condensation during apoptosis and functions as a stress-activated, pro-apoptotic kinase. Upon caspase cleavage, it translocates into the nucleus, triggering DNA fragmentation.
STK4 is a key component of the Hippo signaling pathway, crucial for organ size control and tumor suppression by limiting proliferation and promoting apoptosis.
The pathway involves a kinase cascade where STK3/MST2 and STK4/MST1, in complex with SAV1, activate LATS1/2, which then phosphorylate YAP1 and WWTR1/TAZ to inhibit their nuclear translocation.
This mechanism represses hepatocyte proliferation, prevents activation of adult liver stem cells, and inhibits tumor formation.
