Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha) is a receptor-type transmembrane glycoprotein expressed on cells of myeloid origin, including granulocytes, dendritic cells (DCs), macrophages, mast cells, and hematopoietic stem cells.
SIRP alpha acts as a substrate for several activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase-coupled signaling pathways.
Binding of SIRP alpha to CD47 leads to phosphorylation of ITIM motifs, recruiting SHP-1 and SHP-2 phosphatases.
This interaction inhibits macrophage phagocytosis and contributes to T cell and DC activation.
SIRP alpha regulates cellular responses to serum, growth factors, insulin, oncogenes, and adhesion, playing roles in physiological and pathological processes.
Cancer cells highly express CD47, activating SIRP alpha and inhibiting macrophage-mediated destruction of tumor cells.
