Target Information
Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene.
It is a key structural component of the extracellular matrix in cartilage, providing resistance to compression. The human protein is approximately 2316 amino acids (~250 kDa) and can exist in multiple isoforms due to alternative splicing.
Aggrecan has a bottlebrush-like structure with chondroitin sulfate and keratan sulfate chains attached to its core. It contains three interglobular domains (G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.
Fragments of aggrecan are released into synovial fluid during all stages of osteoarthritis due to cleavage by aggrecanase and MMPs.
Antibodies BC-3 and BC-4 can detect neo-epitopes generated by these enzymes (ARGSVXXX and XXXVDIPEN, respectively).
