Target Information

Functions as a key mediator in apoptosis and inflammation. Promotes caspase-mediated apoptosis involving caspase-8 and caspase-9 in a cell type-specific manner. Involved in activation of the mitochondrial apoptotic pathway and caspase-8-dependent maturation of BID. Mediates mitochondrial translocation of BAX and activates BAX-dependent apoptosis coupled with caspase-9, -2, and -3 activation.

Also involved in macrophage pyroptosis, a caspase-1-dependent inflammatory cell death. Acts as an integral adapter in inflammasome assembly, activating caspase-1 and processing proinflammatory cytokines. Required for recruitment of caspase-1 to inflammasomes containing pattern recognition receptors such as NLRP2, NLRP3, AIM2, and probably IFI16. Facilitates processing of procaspase-1 in NLRP1 and NLRC4 inflammasomes.

In cooperation with NOD2, participates in inflammasome activation by bacterial muramyl dipeptide. May be involved in DDX58-triggered proinflammatory responses. Isoform 2 may regulate inflammasome adapter function, while isoform 3 inhibits IL-1β maturation. Also involved in caspase-1-independent cell death with AIM2 and in dendritic cell maturation for T-cell immunity. Regulates NF-κB signaling by modulating IKK complex activity and RIPK2 association.
