Target Information

MYLK (MLCK) is a serine/threonine kinase that phosphorylates myosin regulatory light chains to assist in the production of contractile activity (myosin–actin interaction). MLCK is responsible for smooth muscle contraction via phosphorylation of a specific serine in the N-terminus of myosin light chains (MLC), facilitating myosin interaction with actin filaments. It plays a central role in vascular permeability and tissue edema formation.
In the nervous system, MYLK regulates astrocytic process growth and participates in transmitter release at synapses between cultured sympathetic ganglion cells. It also contributes to signaling pathways leading to fibroblast apoptosis.
Smooth muscle and non-muscle isozymes are expressed in various adult and fetal tissues, with non-muscle isoform 2 being the dominant splice variant. The Telokin isoform, which binds calmodulin, is also broadly expressed. MYLK is likely down-regulated by phosphorylation and contains one fibronectin type III domain and nine immunoglobulin-like C2-type domains.
