Target Information
GAD-65와 GAD-67은 group II decarboxylase 단백질군에 속하며, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 과정의 속도 결정 단계(rate-limiting step)를 촉매합니다.
두 GAD 모두 뇌에서 발견되지만, GAD-65는 신경 종말의 시냅스 소포막에 국재화되어 있고 GAD-67은 세포 전체에 분포합니다.
GAD-67은 기본적인 GABA 합성에 관여하며, 신경전달에서 GABA 수요가 증가할 때 GAD-65가 일시적으로 활성화되어 GABA 생산을 돕습니다.
GAD-65의 결손은 GABA 신경전달에 장애를 초래할 수 있으며, GAD-67의 결손은 치명적입니다.
GAD-67은 대체 스플라이싱으로 인해 두 가지 isoform(GAD-67, GAD-25)을 가집니다.
