Target Information
Adrenergic receptors (ARs) are members of the 7-transmembrane domain G-protein-coupled receptor superfamily that bind the endogenous catecholamines epinephrine and norepinephrine. Pharmacological, structural, and molecular cloning data indicate significant heterogeneity within this receptor family.
Nine receptor subtypes have been identified, including:
- Alpha-1 AR subtypes: 1A/D, 1B, 1C
- Alpha-2 AR subtypes: 2A, 2B, 2C
- Beta AR subtypes: 1, 2, 3
ARs are involved in various physiological and pathological processes such as hypertension, cardiac dysfunction, diabetes, glaucoma, depression, and impotence.
Alpha-1A adrenergic receptor (A1AR) subtypes are found in numerous tissues and play a role in regulating blood pressure, vascular tone, and cardiac output. A1AR activation influences uterine contraction, hepatic glucose metabolism, HSP70 expression, proto-oncogene activation, and mitogenesis. Additionally, norepinephrine can increase nitric oxide synthase (NOS) levels in the hypothalamus via alpha-1 AR activation, indirectly affecting hormone release (LH, androgens, estrogen, progesterone).
